AMAZE A multicenter trial using Atacand®-Zestril®* versus Zestril to evaluate the effects on lowering blood pressure [*Atacand as add-on therapy with Zestril] AstraZeneca completed a clinical study examining the angiotensin-II receptor blocker, Atacand (candesartan cilexetil) as an add-on therapy with the ACE inhibitor, Zestril (lisinopril), for lowering BP (blood pressure). AMAZE (A Multi-center trial using Atacand-Zestril versus Zestril alone to Evaluate the effects on lowering BP) consists of two parallel, eight-week, multi-centre, double-blind, randomized, parallel-group, forced titration studies involving approximately 1000 hypertensive patients from 10 sites across the U.S. (DSI was one of those sites.) The trial is investigating the effectiveness of adding candesartan to lisinopril, compared to increasing the dose of lisinopril alone, for lowering BP. After an initial open-label treatment period with lisinopril once daily, patients whose BP remained uncontrolled were randomized into one of two treatment groups. The first group was treated with lisinopril + candesartan once daily as add-on therapy. The second group was treated with double the initial dose of lisinopril monotherapy once daily. The design of AMAZE was derived from several studies that examined the BP lowering of candesartan as add-on therapy to an ACE inhibitor. Findings from ACTION, an open-label, large scale, clinical-experience study, showed that candesartan16 to 32 mgm/day as add-on therapy further reduced systolic and diastolic BP in patients already receiving an ACE inhibitor. As one of the principal investigators in AMAZE, AstraZeneca has told DSI that we are receiving an honorable mention in the journal article.
